چکیده

Over-expression of the fibroblast elastase neprilysin (NPL) plays a pivotal role in impairing three-dimensional architecture dermal elastic fibers, which results loss skin elasticity and subsequent wrinkle formation (WF). The expression level NPL human fibroblasts (HDFs) is closely associated with epithelial-mesenchymal cytokine interactions (EMCI) via IL-1α GM-CSF, both have potential to stimulate by HDFs. Since we recently found that topical application hydrogenated retinol (HR) significantly improved wrinkling symptoms facial clinical study, characterized mode action HR association mechanism. When HDFs were co-cultured UVB-exposed primary keratinocytes (HPKs), abrogated increased mRNA levels Analyses cytokines revealed while co-culture stimulated secretion IL-1α, GM-CSF IL-6, those three cytokines. While but not or IL-6 HPKs, conditioned medium at activity protein levels, was attenuated post-UVB treatment HR. there no direct inhibitory effect on activity, these suggest could interrupt EMCI IL-1a abrogate On other hand, IL-1α-treated HDFs, abolished GM-CSF. Taken together, indicate abrogating attributable its UVB-increased HPKs GM-CSF-stimulated Our be an effective agent ameliorate WF down-regulation EMCI.

NDRG2 Regulates the Expression of Genes Involved in Epithelial Mesenchymal Transition of Prostate Cancer Cells

Background: Metastasis is the main cause of prostate cancer (PCa) death. The inhibitory effect of N-myc downstream-regulated gene 2 (NDRG2) on the invasiveness properties of PCa cells has been demonstrated previously. However, its underlying mechanisms have not yet been investigated. The present study aimed to investigate the effects of NDRG2 overexpression on the expression of genes involved i...

Interleukin-6 (IL-6) gene expression and secretion by cytokine-stimulated human retinal pigment epithelial cells.

Retinal and choroidal inflammatory lesions are important causes of visual loss, but the mechanisms regulating intraocular inflammation remain poorly understood. By virtue of its position at the blood-retina barrier, the retinal pigment epithelium (RPE) cells may be critical to the initiation and propagation of ocular inflammation. Previously we showed that cytokine-stimulated RPE cells produce ...

Epithelial-mesenchymal Interactions during Odontogenesis

Recently we reported ultrastructural observatlons which indicated that matrix vesicles were actual constituents of the intercellular organic matrix during embryonic tooth development (14). It is assumed that many of these matrix vesicles are involved in formative aspects of dentinogenesis or amelogenesis, two primary extracellular processes that occur during embryonic tooth formation The fracti...

Nitric oxide attenuates epithelial-mesenchymal transition in alveolar epithelial cells.

Patients with interstitial lung diseases, such as idiopathic pulmonary fibrosis (IPF) and bronchopulmonary dysplasia (BPD), suffer from lung fibrosis secondary to myofibroblast-mediated excessive ECM deposition and destruction of lung architecture. Transforming growth factor (TGF)-beta1 induces epithelial-mesenchymal transition (EMT) of alveolar epithelial cells (AEC) to myofibroblasts both in ...
